Sperm cryopreservation is the process of freezing sperm cells to preserve their viability for future use in fertility treatment. It is a technique that has been practiced clinically since the 1950s, but the sophistication of the process has advanced considerably. Understanding what sperm cryopreservation involves at a cellular level is important not just for clinicians but for anyone responsible for selecting the equipment and protocols that govern it.

At the cellular level, sperm cryopreservation works by slowing biological processes to a point where cell metabolism essentially stops. This is achieved by reducing the temperature of the sperm sample to cryogenic levels, typically stored in liquid nitrogen at minus 196 degrees Celsius. Before freezing, a cryoprotective agent is added to the sample. The most common is glycerol, which works by replacing intracellular water and preventing the formation of ice crystals that would otherwise rupture the cell membrane.

The freezing rate during sperm cryopreservation matters significantly. Too fast and you get ice crystal damage. Too slow and solute toxicity from the cryoprotectant becomes a problem. Most modern protocols use a controlled rate freezer that brings the sample down at a precise speed, often one to five degrees Celsius per minute, before the sample is plunged into liquid nitrogen for long-term storage. The equipment used in this step is not interchangeable. Cheap or poorly calibrated equipment produces inconsistent cooling curves that directly impact post-thaw motility rates.

Long-term storage after sperm cryopreservation is where the storage vessel becomes critical. Samples held at consistent liquid nitrogen temperatures are theoretically stable indefinitely. There is documented evidence of successful fertilisation from sperm stored for over 20 years. But that stability is contingent on the vessel maintaining temperature without fluctuation, which is why the quality of your liquid nitrogen storage dewar directly determines the long-term success of your sperm cryopreservation programme.
