A holding pipette is a micromanipulation instrument used to immobilise an oocyte or embryo on the micromanipulation stage during ICSI, embryo biopsy, or assisted hatching. It works by applying gentle, controlled negative pressure to the zona surface of the oocyte or embryo, creating a stable grip that holds the sample in a fixed position and plane of focus whilst the procedure is performed.
The straight-cut tip geometry creates a flat contact surface against the zona, which allows the pipette to grip without indenting or distorting the zona beyond the suction point. The internal diameter is sized to hold the oocyte or embryo securely — sufficient negative pressure to prevent drift under the injection or biopsy pipette, but not so much that the zona is compressed or the sample is drawn into the pipette. In ICSI, the holding pipette stabilises the oocyte whilst the angled injection pipette penetrates the zona and oolemma. In biopsy procedures, it stabilises the blastocyst or cleavage stage embryo during the aspiration step.
Correct holding pipette positioning — with the sample held at the 9 o’clock position and the polar body at 6 or 12 o’clock — is a basic requirement of ICSI technique and affects fertilisation outcomes.
