A cryovial is a small, threaded, sealable tube manufactured for the storage of biological samples at cryogenic temperatures, typically in or above liquid nitrogen at -195.8 degrees C. Cryovials are used across IVF, sperm banking, cell line banking, biobanking, tissue storage, and research applications wherever a larger-volume, individually identifiable sample container is required.

How do cryovials differ from straws used in IVF?

Straws – 0.3ml or 0.5ml – are the clinical standard for embryo and sperm cryopreservation in IVF, designed for loading into warming devices and compatible with vitrification and slow-freeze protocols. Cryovials offer larger volumes (typically 1.8ml or 2ml) and a different format: a screw-capped tube that can be labelled directly and stored in a cryogenic storage box within a liquid nitrogen vessel. They are more commonly used for sperm banking where larger volumes are stored, for research cell lines, and for biological material that does not require processing through a clinical warming device.

What should cryovials be made from?

Cryovials must be manufactured from materials that remain stable at -195.8 degrees C without cracking, shattering, or losing seal integrity. Polypropylene is the standard material. The thread and seal design is important: externally threaded cryovials are preferred for liquid nitrogen storage as they prevent liquid nitrogen from entering the thread and creating a pressure hazard on warming. The seal must be gas-tight to prevent contamination and to protect sample integrity over extended storage periods.

What storage format do cryovials use?

Cryovials are stored in cryogenic storage boxes, typically in 9×9 (81-well) or 10×10 (100-well) grid formats, sitting inside canisters within a liquid nitrogen storage vessel. Each position in the grid corresponds to a specific sample, enabling rapid location and retrieval. Colour-coded boxes and alphanumeric grids support the documentation and traceability requirements of HFEA-licensed settings and ISO-certified biobanks.

Cryovial integrity at -195.8 degrees C is not a given. A vial that appears undamaged on the shelf can fail in the vessel if the material specification is inadequate. Always verify that consumables are rated for liquid nitrogen immersion, not simply for -80 degrees C freezer use.
